Abstract: Objective To establish a EOGT1 gene knockout rats model for elucidating the function of EOGT1 in
liver injury in future investigation. Methods EOGT1 +/- rats were got by the TALEN gene knockout technology.
EOGT1 -/- rats were obtained after the hybridization of EOGT1 +/- rats. The genotype was identified by PCR
and DNA sequencing technology. Results The genotype identification method was established. There was no
significant difference on the appearance and behavior of all genotype rats. The average weight of EOGT1 -/- rats
were lower than those of EOGT1 +/+ rats 20 days after birth (t = 2.257, P =0.0343) and the reproductive rate of
EOGT1 -/- rats decreased. Conclusion EOGT1 -/- rats were prepared. Compared with EOGT1 +/+ rats, the average
weight and reproductive rate of EOGT1 -/- rats decreased significantly. The construction of gene knockout model
laid the foundation for the study of the effects of O-GlcNAc on Notch glycosylation in liver injury.
