Gab1蛋白在肝细胞癌组织中的表达及与临床预后的关系

摘要:目的 探讨生长因子受体相关结合蛋白1(growth factor receptor binding protein-associated binder1, Gab1)蛋白在肝细胞癌(hepatocellular carcinoma,HCC)组织中的表达及对患者临床预后的影响。方法 收集2005年1月至2014年2月邯郸市中心医院存档的70例行根治性手术的HCC患者资料,所有患者均未进 行过相关放射治疗或化学治疗。同时选取不典型增生和正常肝组织各30例为研究对象。采用免疫组织 化学(EnVision TM )法检测Gab1蛋白在上述3种组织中的表达,并分析其与HCC临床病理因素及HCC患 者4年生存率的关系。结果 Gab1蛋白在正常肝组织、不典型增生及HCC组织中阳性表达率分别为0.00% (0/30)、33.33%(10/30)和58.57%(41/70),差异有统计学意义(χ 2 = 30.78,P < 0.001)。Gab1 蛋白的阳性表达率与患者年龄、性别、组织分型及病灶形式无关(P > 0.05),但与肿瘤直径、淋巴 结转移、MCM7蛋白表达及临床TNM分期有关(P < 0.05)。Gab1阳性表达的HCC患者4年生存率为 5.26%,阴性表达者为25.00%,差异有统计学意义(χ 2 = 3.96,P = 0.04)。结论 Gab1蛋白的过表达参与 了HCC的发生发展,其表达水平可作为预测HCC患者4年生存预后的参考指标。
Abstract: Objective To investigate the expression of growth factor receptor binding protein-associated binder1 (Gab1) protein in hepatocellular carcinoma (HCC) tissue and its influence on clinical prognosis of HCC. Methods The clinical data of 70 patients with HCC who underwent radical surgery in Central Hospital of Handan from January 2005 to February 2014 were collected, and all patients had no relevant radiotherapy or chemical treatment. Another 30 cases of atypical hyperplasia and 30 cases of normal liver tissue were also selected as the subjects. The expression of Gab1 protein in HCC tissue, atypical hyperplasia and normal liver tissues were detected by immunohistochemistry (EnVision TM ). The relationship between the expression of Gab1 protein and clinicopathological factors of HCC and 4-year survival rate were analyzed. Results The positive expression rates of Gab1 protein in normal tissue, atypical hyperplasia and HCC tissue were 0.00% (0/30), 33.33% (10/30) and 58.57% (41/70), respectively. The difference was statistically significant (χ 2 = 30.78, P < 0.001). The positive expression of Gab1 protein in HCC tissue had no relationship with age, gender, histological type and lesion formation (P > 0.05), but was related to tumor diameter, lymphaden metastasis, MCM7 protein expression and clinical TNM stage (P < 0.05). The 4-year survival rate of HCC patients with positive expression of Gab1 protein was 5.26% and 25.00% in negative expression patients. The difference was statistically significant (χ 2 = 3.96, P < 0.05). Conclusions The overexpression of Gab1 protein is involved in the development of HCC, and its expression level can be used as a prognostic indicator for predicting the 4-year survival of patients.
