Objective To detect gene expression of prohibitin in human liver cancer cells SMMC7721 and Huh-7, and amplify the full coding sequence of prohibitin. Methods The total cellular RNA of SMMC7721 and Huh-7 were extracted, then the first strand cDNA was synthesized by reverse transcription. The expression of prohibitin mRNA was verified by Real time PCR, with GAPDH as an internal validation. RT-PCR was used to amplify the full coding sequence of prohibitin. Results The results of Real time PCR showed prohibitin was expressed in both SMMC7721 and Huh-7 cells. And the expression level of prohibitin in SMMC7721 cells was 6.652 times higher than that in Huh-7 cells. About 835 bp full coding sequence of prohibitin was successfully amplified by RT-PCR, without unspecific amplificaiton. Conclusions The expression level of prohibitin in human liver cancer cells SMMC7721 was relatively high.
